If you're configuring Hybrid Deployment, your minimum downtime for migrating email from your local server to Exchange Online is no downtime at all. In a Hybrid Deployment you're using Exchange 2010 functionality to make mailbox move requests, rather than migrate the mailboxes. As long as you're coming from an Exchange 2007 or Exchange 2010 environment, a mailbox move request keeps the mailbox online throughout the move, allowing the user to continue to send and receive mail with it. Once fully moved, it prompts the user to sign out and sign back into their account, after which they'll be completely moved over and will have experienced no downtime at all.
