We are being migrated from BPOS to Office 365 this weekend. Do we need to amend the settings on our users iPhones in order for them to continue accessing their email? Or will they work ok once the migration is complete?

Yes, you need to change the settings. If you have a mobile device that connected to BPOS, you need to set it up to connect to Office 365. For an iPhone, see Access Your Account Using a Mac.
